Description:
The Unversity of Ottawa is pleased to invite qualified proponents to submit a proposal for a Cryo-holder system for transmission electron microscopy. The requested cryo-transfer holder system is a critical piece of infrastructure in a drug-discovery pipeline that is being developed at the University of Ottawa. The cryo-transfer holder system will facilitate the screening of vitrified protein sample grids on a JEM-1400Plus 120 kv transmission electron microscope (TEM) for their suitability for further imaging on a high-end (200 or 300 kv) microscope designed for single particle analysis. The goal is to identify grids that are suitable for solving protein 3D structures. General Description: The cryo-transfer holder system allows the operator to first prepare and then image vitrified TEM grids at cryo-temperatures on a TEM - in this case the JEM-1400Plus 120 kv TEM that is already in use at uOttawa. The cryo-transfer holder system includes the cryo-transfer holder itself, which maintains cryo-temperatures during imaging on the microscope, a temperature controller, a workstation that facilitates the transfer, under liquid nitrogen, of vitrified grids from a sample storage box to the cryo-transfer holder, and all accessory devices, such as a vacuum pumping unit and/or vacuum storage container, etc., required to maintain the cryo-transfer holder in functional working condition.
